BIO:ON Insight EN #4 · Jun 16, 2026 · Biosimilar · Bispecific Antibody · New Target Copy, combine, or go where no drug has gone:
|
|
|
Putting this week's news together, one question came to mind: which direction is drug innovation actually heading? On one side, a biosimilar makes a proven drug cheaper. In the middle, a bispecific antibody ties two known targets into one molecule. And at the far end, a company takes its first shot at a target no one had ever turned into a drug. Three very different routes, all trying to reach patients.

🔤 Word of the Week
Bispecific Antibody (PD-L1×VEGF)
The mechanism behind this week's BioNTech and BMS story (Story 2)
A normal antibody grabs one target. A bispecific antibody has two different "claws" on one molecule, so it can grab two targets at once. This week's pumitamig binds the immune checkpoint protein PD-L1 and the blood-vessel growth factor VEGF-A. It releases the immune brake (PD-L1) and tidies up tumour blood vessels (VEGF-A), effectively two drugs in one.
Arm 1: PD-L1 — release the immune brake
Tumours display PD-L1 to switch off attacking T cells. Blocking it lets immune cells go back on the attack. It's the same axis drugs like Keytruda target.
Arm 2: VEGF-A — normalise tumour vessels
Tumours use VEGF-A to build messy blood vessels that keep immune cells out. Blocking it tidies up the vessels so immune cells can get in. Together: brake off, and a clear road in.
📰 This Week's Top 3 Stories
Bio-Thera Wins FDA Approval for the First Biosimilars to Simponi and Simponi Aria
Bio-Thera Solutions press release · US partner Accord BioPharma | Launched May 18, 2026
Key Facts
The FDA approved two golimumab biosimilars developed by Bio-Thera Solutions. They're the first biosimilars referencing J&J's Simponi (subcutaneous) and Simponi Aria (intravenous). Golimumab is an anti-TNFα monoclonal antibody used in rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis and ulcerative colitis. Accord BioPharma handles US commercialisation and launched on May 18, 2026.
Plain-English Breakdown 🧩
A biosimilar is a copy of an off-patent biologic that's been shown to be just as safe and effective. Unlike small-molecule generics, biologics are made in living cells, so a biosimilar isn't identical, but the FDA has judged there's no clinically meaningful difference. The key point: Simponi had no biosimilar competition until now. This is the first challenger, which is where lower prices and wider access start.
🌍 Global Lens
Bio-Thera is a Chinese company, and this approval is part of a bigger shift: Asian biosimilar makers such as Celltrion and Samsung Bioepis are now major suppliers to Western markets. Europe usually adopts biosimilars faster than the US, because national tenders and hospital purchasing push prices down quickly, so the US launch will be an interesting test of how fast a first challenger can take share.
Why it matters for your career: Biosimilar companies hire heavily in regulatory, manufacturing and market access. Knowing that first-mover advantage and supply matter as much as the science goes a long way in an interview.

BioNTech and BMS's PD-L1×VEGF Bispecific pumitamig Shows Strong Responses in First-Line Lung Cancer
BioNTech (Nasdaq: BNTX) · Bristol Myers Squibb (NYSE: BMY) press release | 2026
Key Facts
BioNTech and Bristol Myers Squibb (BMS) released interim Phase 2 data from the global Phase 2/3 ROSETTA Lung-02 trial in previously untreated advanced non-small cell lung cancer (NSCLC). Patients received pumitamig, a bispecific that hits PD-L1 and VEGF-A, plus chemotherapy. The companies reported encouraging anti-tumour activity and high response rates in both squamous and non-squamous NSCLC, across all PD-L1 expression levels. Specific response and survival numbers haven't been released yet.
Plain-English Breakdown 🧩
The usual first-line treatment for lung cancer is an immunotherapy (PD-1/PD-L1) plus chemotherapy. pumitamig adds VEGF blocking in the same molecule. Patients with low PD-L1 expression usually get less benefit from immunotherapy, so responses across all expression levels suggest it could help a wider group. It's still interim Phase 2, though, with Phase 3 to come. The Word of the Week above explains the mechanism.
🌍 Global Lens
The PD-(L)1×VEGF idea took off in China. Akeso's ivonescimab showed it could work first, and BioNTech's own molecule came from Biotheus, a Chinese biotech it bought. So this is a Western pairing competing on a mechanism China pioneered, and Akeso's US partner Summit is running the same race. Where the first global approvals land will say a lot about who leads oncology innovation now.
Why it matters for your career: Clinical and medical teams want people who can be excited about a result and still say clearly that an interim readout isn't proof. That balance is harder than it sounds.

Orion Reports First-in-Human Results for ODM-212, an Oral pan-TEAD Inhibitor
Orion Pharma (HEL: ORNBV) press release · TEADES Phase 1/2 | 2026
Key Facts
Orion Pharma reported the first Phase 1 results for ODM-212 from its ongoing first-in-human Phase 1/2 TEADES trial in patients with advanced solid tumours. ODM-212 is an oral pan-TEAD inhibitor. TEAD proteins are the key transcription factors of the Hippo pathway; together with YAP/TAZ they drive tumour growth and drug resistance, and they've long been considered hard to drug. ODM-212 blocks the whole TEAD family at once. Specific efficacy and safety numbers haven't been released yet.
Plain-English Breakdown 🧩
Unlike a biosimilar (copying a proven drug) or a bispecific (combining proven targets), ODM-212 is a first attempt at a target nobody had turned into a drug. Programmes like this have lower odds of success, but when they work they open a market with no competitors: high risk, high reward. "First Phase 1 results" means they've only just started checking whether it can be given safely to people.
🌍 Global Lens
Orion is Finnish, which is a nice reminder that first-in-class science doesn't only come from US biotechs or the biggest pharma companies. Mid-sized European firms can't outspend the giants, so they often pick hard, unfashionable targets where being early matters more than being big. Several other companies are also chasing TEAD, so this is a global race that's barely started.
Why it matters for your career: Early development teams value people who can explain why a target matters without overselling a first-in-human readout.

📈 3 Stocks That Moved This Week
Direction and catalyst, sketched — not exact prices. Check a real chart for that.
|
BioNTech NASDAQ: BNTX |
|
▲ Strong |
Positive interim Phase 2 lung cancer data for pumitamig lifted sentiment, as investors bought into the combine strategy. Hiring signal: a bispecific heading into Phase 3 usually means more clinical operations and translational roles in oncology. (Directional sketch.)
|
Bristol Myers Squibb NYSE: BMY |
|
▲ Edging up |
Benefited as pumitamig's partner, but as a large cap it moved more gently than BioNTech. (Directional sketch.)
|
Orion Pharma HEL: ORNBV |
|
≈ Flat |
With ODM-212 at first Phase 1 results, the market is waiting and seeing. Orion's larger businesses (Parkinson's and prostate cancer) mean a new-target story barely moves the stock. (Directional sketch.)
🔎 Why this pattern
The size of each reaction matched how far along the data was. BioNTech, with clinical data, rose clearly; its large partner BMS edged up; Orion, at first Phase 1 results, barely moved. Same week of good news, but the market priced it by data maturity. (All directional sketches — check a real chart for exact prices.)
